Illuminating the lineage-specific diversification of resin glycoside acylsugars in the morning glory (Convolvulaceae) family using computational metabolomics

Lars H. Kruse et al.

Summary: In this study, the structural diversity of resin glycosides (RGs) in different species of the morning glory family was analyzed using a phylogeny-guided approach. The researchers discovered substantial lineage-specific differentiation in the utilization of sugars, hydroxyacyl chains, and decorating acyl chains, especially in the two largest genera in the family. They also developed a computational workflow that explained a significant portion of the MS/MS fragments and predicted the structural components of previously characterized RGs. Overall, this study improves our understanding of phytochemical diversity and lays a foundation for characterizing the evolutionary mechanisms underlying RG diversification.

Identification and characterization of the key enzyme in the biosynthesis of the neurotoxin β-ODAP in grass pea

Moshe Goldsmith et al.

Summary: Grass pea is a commonly grown grain legume with high nutritional value. However, it produces a neurotoxic compound that can cause severe neurological disorders. In this study, the enzyme responsible for the formation of this compound, beta-ODAP synthase (BOS), was identified and characterized. The expression of BOS in the presence of its substrates was found to be sufficient for beta-ODAP production. This discovery may help in the development of grass pea cultivars that are safe for consumption.

Orthology-based analysis helps map evolutionary diversification and predict substrate class use of BAHD acyltransferases

Lars H. Kruse et al.

Summary: Large enzyme families catalyze metabolic diversification by using diverse chemical scaffolds, but it is not clear how these families achieve functional diversity. In this study, using BAHD acyltransferases as a model, the researchers compiled published activities and used orthologous groups to predict the utilization and origins of BAHD enzymes in plants. The results not only describe how BAHD enzymes contributed to the evolution of multiple chemical phenotypes in the plant world, but also propose an approach for improved functional annotation of plant enzyme families using biocuration.

Identification of BAHD acyltransferases associated with acylinositol biosynthesis in Solanum quitoense (naranjilla)

Bryan J. Leong et al.

Summary: Plants produce specialized metabolites that play important roles in their interactions with animals, microbes, and other plants. This study focuses on acylsugars, protective metabolites found in the trichomes of Solanaceae family plants. The researchers characterized enzymes involved in acylinositol biosynthesis in the Andean fruit plant Solanum quitoense and found that changes in gene expression led to changes in acylinositol accumulation. They also discovered that pH can influence the stability and rearrangement of the biosynthesis product. Surprisingly, the in vitro products of these enzymes were different from those found in planta, indicating that additional enzymes are required for acylinositol biosynthesis.
