Endothelial Cells Promote Osteogenesis by Establishing a Functional and Metabolic Coupling With Human Mesenchymal Stem Cells

Summary: This study reveals the molecular mechanism underlying the interaction between endothelial cells and osteodifferentiating stem cells during bone tissue renewal. Specifically, endothelial cells induce osteodifferentiation in stem cells through the secretion of soluble factors, while stem cells promote vascular network formation by recruiting endothelial cells. Moreover, endothelial cells enhance anaerobic glycolysis in stem cells without compromising their oxidative metabolic shift required for osteodifferentiation.

Endothelial Heme Dynamics Drive Cancer Cell Metabolism by Shaping the Tumor Microenvironment

Summary: The communication between cancer cells (CCs) and stromal cells in the tumor microenvironment (TME) is crucial for cancer progression. In this context, the importance of endothelial cells (ECs) lies in their vascular functions in supporting tumor growth. Targeting endothelial heme metabolism by interfering with the cell membrane heme exporter FLVCR1a in tumor-associated ECs (TECs) can affect fatty acid oxidation (FAO) and induce ketone bodies (KBs) accumulation, leading to a metabolic rewiring in cancer cells. This study suggests new metabolic vulnerabilities for potential cancer therapy.

The heme synthesis-export system regulates the tricarboxylic acid cycle flux and oxidative phosphorylation

Summary: There is a functional axis between heme synthesis and heme export, with heme efflux through the plasma membrane sustaining heme synthesis and modulating tricarboxylic acid cycle and oxidative phosphorylation. Inhibition of heme export reduces heme synthesis and promotes TCA cycle fueling and flux as well as oxidative phosphorylation. These findings suggest that the heme synthesis-export system modulates TCA cycle and oxidative metabolism, providing a mechanistic basis for their enhancement in cells with high-energy demand.
