Summary: In this study, BDTR9-OC8 and BDTR9-C8 nanoparticles with different side chain rigidities were investigated for their phototheranostic properties. It was found that BDTR9-C8 nanoparticles exhibited enhanced NIR-II FLI and PAI brightness compared to BDTR9-OC8 nanoparticles. The enhanced signals were attributed to the amorphous condensed state of BDTR9-C8 nanoparticles, which facilitated the transportation of photo-generated excitons and phonons. Additionally, both nanoparticles showed excellent photothermal conversion efficiency and have potential applications in photothermal therapy.

Encapsulating BH 1024 into folate-functionalized polymer demonstrated a synergetic PDT/PTT effect in vitro and in vivo, eventually achieving solid tumors elimination under NIR-II fluorescence guide. This is the first attempt to explore and design small molecular dyes for NIR-II PDT or NIR-II PDT/PTT.
