Peroxide-Mediated Oxygenation of Organic Compounds by Fungal Peroxygenases

Martin Hofrichter et al.

Summary: Unspecific peroxygenases are fascinating biocatalysts that efficiently transfer oxygen from peroxide to a wide range of organic substrates, combining peroxidase and monooxygenase activities. They have been studied for nearly 20 years and show great potential in biotechnology.

Surfing the wave of oxyfunctionalization chemistry by engineering fungal unspecific peroxygenases

Alejandro Beltran-Nogal et al.

Summary: The selective insertion of oxygen into non-activated organic molecules is crucial for the synthesis of existing and next generation industrial chemicals or pharmaceuticals. Fungal unspecific peroxygenases (UPOs) are highly active biocatalysts for C-H oxyfunctionalization, but their limited availability has hindered their use in industry. However, the combination of directed evolution and computational design is bringing UPOs closer to practical applications.

Recent developments in the use of peroxygenases - Exploring their high potential in selective oxyfunctionalisations

Markus Hobisch et al.

Summary: Peroxygenases are a new class of enzymes that allow selective oxyfunctionalisation reactions without the need for a cofactor, different from well-known P450 monooxygenases. Recent developments in organic synthesis, molecular biotechnology, and reaction engineering have focused on utilizing these enzymes in industrial applications. There is great potential for peroxygenases in selective oxyfunctionalisations and further studies are required to explore their full capabilities in organic synthesis.

Advances in enzymatic oxyfunctionalization of aliphatic compounds

Carmen Aranda et al.

Summary: Selective oxyfunctionalizations of aliphatic compounds are challenging chemical reactions, but enzymes such as P450 monooxygenases and unspecific peroxygenases (UPOs) play important roles due to their stereo- and regio-selectivity. UPOs, discovered in fungi, have emerged as promising biocatalysts for their ability to catalyze the oxyfunctionalization of aliphatic and aromatic compounds using only H2O2.

Enzymatic Hydroxylations of sp3-Carbons

Judith Muench et al.

Summary: The enzymatic hydroxylation of activated and nonactivated sp(3)-carbons is challenging in organic synthesis, but nature provides a variety of enzymes with catalytic versatility to tackle this task. Different enzymes have distinct specificity in substrate scope, selectivity, activity, stability, and catalytic cycle, and factors such as heterologous production, crystal structure availability, enzyme engineering potential, and substrate promiscuity play crucial roles in the applicability of these biocatalysts.

Identification and Expression of New Unspecific Peroxygenases - Recent Advances, Challenges and Opportunities

Alina Kinner et al.

Summary: The fungal heme-thiolate enzyme subfamily of unspecific peroxygenases (UPOs) was first described in 2004 and has a wide range of oxidative transformation applications. However, their availability for potential biotechnological applications is restricted by challenges in identifying new natural biocatalysts, their production, and describing their properties.

Summary: The study reveals that the peroxidative activity of UPOs is not dependent on a long-range electron transfer pathway from oxidizable residues at the protein surface, unlike other ligninolytic peroxidases. Rather, it seems to be mainly located at the heme access channel.
