Rationally Designed Two-Photon Ratiometric Elastase Probe for Investigating Inflammatory Bowel Disease

Summary: By developing a fluorescent elastase probe, we have investigated elastase level changes in various mouse organs under an IBD condition for the first time. The results reveal significantly higher elastase levels in the liver and duodenum of healthy mice than in other organs. Under the IBD condition, elastase level changes were observed in the liver, duodenum, colon, and lung. The probe offers an ideal tool for mapping neutrophil migration in the body.

Essential Loop Dynamics Modulates Catalytic Activity in α-Chymotrypsin

Summary: This study investigates the conformational dynamics and catalytic activity of alpha-chymotrypsin using optical spectroscopy and molecular dynamics simulations. The enzyme shows highest catalytic efficiency at the optimum temperature of 308 K. It is found that the flexibility of the enzyme increases with temperature. Principal component analysis reveals the essential dynamics and gatekeeping role of loop regions surrounding the catalytic pocket.

De novo design of an ultrasensitive fluorogenic probe for aminopeptidase N sensing in living system

Summary: Aminopeptidase N (APN) is an important metalloprotease that is widely expressed and plays a significant role in maintaining normal physiological functions and the progression of chronic inflammatory diseases and cancer. This research proposes a new design strategy using an unnatural amino acid, 2-Aminobutyric acid (Abu), to construct a near-infrared fluorescent probe TMN-Abu for sensitive and specific detection of APN activity. The probe showed high sensitivity and binding affinity towards APN, and successfully monitored APN activity in living cells, tumor-bearing mice, diabetic model mice, and biological events such as starvation and EGF stimulation.
