Real-Time Monitoring of Neurotransmitters in the Brain of Living Animals

Highly Stable Electrochemical Probe with Bidentate Thiols for Ratiometric Monitoring of Endogenous Polysulfide in Living Mouse Brains

Summary: A lack of reliable approaches for real-time measurement and quantification of polysulfides in vivo has limited exploration of their roles in brain functions. In this study, an electrochemical probe was designed for determination of H2Sn, showing high selectivity for real-time tracking in a linear range of 0.25-20 μM. The developed microelectrode with high selectivity, accuracy, and stability was successfully employed for in vivo assaying of H2Sn in mouse brains with ischemia.

Raman Fiber Photometry for Understanding Mitochondrial Superoxide Burst and Extracellular Calcium Ion Influx upon Acute Hypoxia in the Brain of Freely Moving Animals

Summary: The development of a novel tool for real-time monitoring and simultaneous quantitation of multiple molecules in mitochondria in freely moving animals is crucial for understanding the roles of mitochondria in brain functions. The study utilized a Raman fiber photometry to track and quantify mitochondrial O-2(.-), Ca2+, and pH in different brain regions, revealing changes in these parameters from superficial to deep brain regions during hypoxia. Furthermore, it was discovered that hypoxia-induced mitochondrial O-2(.-) burst was regulated by ASIC1a, leading to mitochondrial Ca2+ overload attributed to the influx of extracellular Ca2+.

Highly sensitive and selective nanostructured microbiosensors for glucose and lactate simultaneous measurements in blood serum and in vivo in brain tissue

Summary: Highly sensitive and selective nanostructured lactate and glucose microbiosensors were developed for simultaneous determination in rat brain. The microbiosensors, based on carbon fiber microelectrodes modified with nanoporous gold, exhibited enhanced sensitivity and electrocatalytic properties towards H2O2 detection. The selective measurements were achieved by enzyme immobilization, permselective layer, and a null sensor. The microbiosensor platform was used to determine lactate and glucose in blood serum samples, as well as in vivo measurements of basal extracellular concentrations in different rat brain structures.

A tissue-like neurotransmitter sensor for the brain and gut

Summary: Neurotransmitters play crucial roles in regulating neural circuit dynamics in the central and peripheral nervous systems, including the gastrointestinal tract. However, there is a lack of bioelectronic tools for real-time monitoring of neurotransmitter dynamics in vivo, especially in the enteric nervous system. In this study, the researchers developed a tissue-mimicking, stretchable neurochemical biological interface called NeuroString, which allows chronic in vivo real-time monitoring of monoamine levels in the brain and serotonin dynamics in the gut. This interface has the potential for studying the impact of neurotransmitters on gut microbes and brain-gut communication.

Efficient Electrochemical Microsensor for In Vivo Monitoring of H2O2 in PD Mouse Brain: Rational Design and Synthesis of Recognition Molecules
