Sustained Coinfections with Staphylococcus aureus and Pseudomonas aeruginosa in Cystic Fibrosis

Anthony J. Fischer et al.

Summary: Staphylococcus aureus and Pseudomonas aeruginosa are both abundant in sputum cultures of cystic fibrosis patients. Contrary to common assumption, there is no rapid replacement of S. aureus by P. aeruginosa. Many CF patients have long-term coinfection with these organisms.

Branched-chain amino acid metabolism controls membrane phospholipid structure in Staphylococcus aureus

Matthew W. Frank et al.

Summary: Branched-chain amino acids, particularly isoleucine, play a crucial role in regulating virulence in Staphylococcus aureus by being converted to precursor molecules for fatty acid synthesis. The selectivity of how the bacterium metabolizes isoleucine influences the composition of membrane phospholipids, demonstrating the importance of isoleucine metabolism in controlling membrane structure.

Application of Thin-Film Microextraction to Analyze Volatile Metabolites in A549 Cancer Cells

Wojciech Filipiak et al.

Summary: Volatile organic compounds (VOCs) have been proposed as biomarkers for disease detection, and a modified SPME technique, thin-film microextraction (TFME), has been suggested for more sensitive analysis of cellular VOCs. Among different adsorbents, HLB sheets showed the most versatility in analyzing VOCs with a broad polarity and volatility. Overall, the study successfully applied the established method to analyze metabolites produced by A549 cells, identifying five volatiles at significantly higher levels in the cell culture medium compared to the cell-free reference medium.

Branched chain fatty acid synthesis drives tissue-specific innate immune response and infection dynamics of Staphylococcus aureus

Xi Chen et al.

Summary: The diverse fatty acid composition in the bacterial cell envelope affects membrane fluidity and immune responses. Shifting away from branched chain fatty acids towards unsaturated fatty acids can disrupt infection dynamics and lead to immune activation. This study highlights the importance of acyl chain repertoire in bacterial infection outcomes and immune responses.
