Getting to HBV cure: The promising paths forward

Scott Fung et al.

Summary: Chronic HBV infection is a global health burden, and achieving a cure remains challenging due to the persistence of cccDNA, HBV-DNA integration, and impaired immune response. Current therapies include DAAs and immunomodulators, but there are still many obstacles to overcome for a complete cure of HBV.

Interferon add-on therapy increased clinical cure significantly for interferon-experienced chronic hepatitis B patients with low HBsAg

Xiaoan Yang et al.

Summary: This study aimed to investigate whether interferon-experienced patients could achieve the same HBsAg clearance and seroconversion rates as interferon-naive patients, and to clarify the associated factors and cut-offs. The results showed satisfactory clinical cure of interferon add-on therapy for interferon-experienced patients with low HBsAg. Age, baseline HBsAg, and week 12 HBsAg were negatively correlated with week 48 HBsAg clearance and seroconversion. Interferon-experienced patients with low HBsAg were suggested with interferon add-on therapy for achieving clinical cure.

Toll-Like Receptor 8 Agonist GS-9688 Induces Sustained Efficacy in the Woodchuck Model of Chronic Hepatitis B

Stephane Daffis et al.

Summary: GS-9688 treatment in woodchucks with WHV infection showed significant antiviral efficacy, with a >5 log(10) reduction in viral load and sustained response even after treatment cessation. The treatment also resulted in reduced intrahepatic WHV RNA and DNA levels, and enhanced immune responses in woodchucks.

Therapeutic Potential of TLR8 Agonist GS-9688 (Selgantolimod) in Chronic Hepatitis B: Remodeling of Antiviral and Regulatory Mediators

Oliver E. Amin et al.

Summary: GS-9688 activates immune cell subsets and induces the production of immunomodulatory mediators in healthy controls and CHB patients. It boosts antiviral effector functions by activating HBV-specific CD8(+) T cells, CD4(+) follicular helper T cells, NK cells, and mucosal-associated invariant T cells. Additionally, GS-9688 modulates immunoregulatory subsets, potentially optimizing its antiviral efficacy.

Toll-Like Receptor Response to Hepatitis B Virus Infection and Potential of TLR Agonists as Immunomodulators for Treating Chronic Hepatitis B: An Overview

Mohammad Enamul Hoque Kayesh et al.

Summary: Chronic hepatitis B virus (HBV) infection is a major global health problem, and the link between Toll-like receptors (TLRs) and chronic HBV infection is being increasingly recognized. TLR agonists have the potential to enhance the host immune response against HBV infection and improve CHB treatment.

Summary: Selgantolimod was found to be safe and well-tolerated in patients with chronic hepatitis B (CHB), inducing cytokine responses and antiviral immunity. Although some patients experienced adverse events and laboratory abnormalities, they were mild or moderate in severity. Further studies with longer treatment durations are needed to assess efficacy.

NK Cell-Derived IL-10 Supports Host Survival during Sepsis

Isaac J. Jensen et al.

Summary: NK cells play a critical role in host survival during sepsis by limiting the scope and duration of the cytokine storm, with NK cell-derived IL-10 being crucial for survival in septic patients. This demonstrates the beneficial regulatory mechanisms of NK cells in controlling systemic inflammation during dysregulated infection-induced inflammatory responses.

Summary: Chronic hepatitis C patients treated with direct acting antivirals (DAA) showed recovery in cell-mediated immune response and TLR8 expression and functionality post-treatment. Additionally, DAA treatment significantly decreased the expression and activation of TLR8 in PBMCs, leading to a reduction in Th1 response levels.
