Stapled Liposomes Enhance Cross-Priming of Radio-Immunotherapy

Yu Zhao et al.

Summary: The development of an antigen-capturing stapled liposome (ACSL) enhances the cross-presentation of tumor-associated antigens in dendritic cells, leading to a strong anti-tumor immune response. In mouse models, ACSL encapsulating l-arginine significantly suppresses tumor growth and induces an abscopal effect. This suggests that ACSL could be a universal platform for the rapid development of various combinations of anticancer therapies using radiosensitizers, photosensitizers, and photothermal agents.

Disrupting N-glycan expression on tumor cells boosts chimeric antigen receptor T cell efficacy against solid malignancies

Beatrice Greco et al.

Summary: In solid tumors, the expression of N-glycans on tumor cells is negatively correlated with the killing ability of CAR T cells. Inhibiting N-glycan synthesis enhances CAR T cell activity and reduces interference from the PD-1-PD-L1 axis. The combination of 2DG and CAR T cell therapy shows effectiveness against multiple carcinomas.

Immune modulating nanoparticles depleting tumor-associated macrophages to enhance immune checkpoint blockade therapy

Zheng Chunxiong et al.

Summary: In this study, an immune modulating nanoparticle (IMNP) was designed to achieve spatial control of drug release sites and synergistically deplete tumor-associated macrophages (TAMs). The results showed that the IMNP effectively eliminated TAMs and improved the tumor environment, leading to a significant enhancement of the anti-tumor efficacy of immune checkpoint blocking therapies.
