Scoping review of the applications of peptide microarrays on the fight against human infections

Arthur Vengesai et al.

Summary: This scoping review explores the use of peptide microarrays in the fight against infectious diseases. The majority of the studies focused on mapping B-cell epitopes and diagnosing infections, while a smaller proportion investigated immunosignature characterization and viral/bacterial binding assays. Peptide microarrays were found to be valuable tools in identifying diagnostic and vaccine targets, as well as antimicrobial peptides.

A systematic and meta-analysis review on the diagnostic accuracy of antibodies in the updates serological diagnosis of COVID-19

Arthur Vengesai et al.

Summary: This study evaluated the diagnostic accuracy of different serological tests for detecting SARS-CoV-2 antigens. Results showed that CLIA and ELISA tests performed better in terms of sensitivity compared to LFIA. A combined IgG/IgM test seemed to be a better choice in terms of sensitivity than measuring either antibody type independently.

SARS-CoV-2 Epitope Mapping on Microarrays Highlights Strong Immune-Response to N Protein Region

Angelo Musico et al.

Summary: A workflow for rapid epitope discovery of SARS-CoV-2 on peptide microarrays was reported, leading to the identification of immunodominant regions on the spike, nucleocapsid, and Orf1ab proteins. The epitope of N protein's region 155-171 showed good diagnostic performance in discriminating Covid-19 positive individuals, with 92% sensitivity and 100% specificity for IgG detection. This epitope represents a promising candidate for further development and rapid implementation in serological tests.

Summary: Antibody tests for SARS-CoV-2 have many uses for public health decisions, but current demand comes mostly from individuals. However, due to uncertainty about the tests and their implications for immunity, positive results should not influence individual decisions, and antibody testing should remain a public health tool.

How to interpret and use COVID-19 serology and immunology tests

David S. Y. Ong et al.

Summary: Molecular tests are considered the gold standard for COVID-19 diagnostics, but serological and immunological tests can also be valuable in certain situations. The presence of antibodies against SARS-CoV-2 may indicate a recent or past COVID-19 infection, and tests like ELISA and CLIA have higher sensitivities. Cellular immunity is crucial in host defences against viruses, and T-cell responses against SARS-CoV-2 can provide important information on past infections.

EpiCurator: an immunoinformatic workflow to predict and prioritize SARS CoV-2 epitopes

Cristina S. Ferreira et al.

Summary: This study introduced the EpiCurator workflow to predict and prioritize epitopes from SARS-CoV-2 genomes, discovering 11 new epitopes in the receptor-binding domain (RBD) with high antigenicity and broad population coverage. The computational validation of a multi-epitope design showed efficiency in triggering an effective immune response. Overall, the findings demonstrate the efficacy of the workflow for epitope discovery, providing potential target candidates for immunogen development.

Summary: This study demonstrated the immunoreactivity of a designed M protein of the SARS-CoV-2 virus with IgM and IgG antibodies in serum samples from COVID-19 convalescents. The ELISA results showed over 93% positivity for IgM detection and over 96% positivity for specific IgG detection to the M protein in COVID-19 sera. Based on its ability to provoke an immune response and strong antigenic properties, the M protein has the potential for diagnostic purposes and vaccine design.
