Collective Plasmon Coupling in Gold Nanoparticle Clusters for Highly Efficient Photothermal Therapy

Summary: This study reports the development of plasmonic nanoparticle clusters (PNCs) as highly efficient photothermal therapy agents (PTAs) and provides a semi-quantitative calculation method for their resonant frequency and absorption efficiency. The optimized gold-based PNCs achieved a photothermal conversion efficiency of up to 84%. The study further demonstrates the extraordinary performance of the optimized PNCs in in vivo applications.

Summary: We demonstrate noninvasive and five-plex SERS imaging of nanoparticle accumulation in tumors of living mice using multispectral palettes of gold multicore-near-infrared (NIR) resonant Raman dyes-silica shell SERS (NIR-SERRS) nanoparticle oligomers. The method allows for ratiometric imaging of tumors by varying the administered ratio of nanoparticles, simulating the detection of multiple biomarkers with different expression levels in the tumor environment. This approach surpasses the multiplicity limit of previous preclinical tumor imaging methods while maintaining sufficient sensitivity for in vivo imaging, enabling noninvasive assessment of multiple biological targets within the tumor microenvironment in living subjects.
