The association of changes of sleep architecture related to donepezil: A systematic review and meta-analysis

Summary: Our meta-analysis found that after treatment with donepezil, the percentage of REM sleep significantly increased in patients, while the percentage of stage 2 sleep, sleep efficiency, and total sleep time decreased. Larger sample size studies are needed to further validate these findings.

Hyperexcitable arousal circuits drive sleep instability during aging

Summary: Sleep quality declines with age and this study identified hyperexcitable hypocretin/orexin (Hcrt/OX) neurons as drivers of sleep fragmentation during aging. Activation of Hcrt neurons increased wakefulness and aged Hcrt neurons showed hyperexcitability and impaired M-current. Disruption of Kcnq2/3 genes in young mice led to sleep destabilization resembling aging-associated sleep fragmentation.
