Drug affinity responsive target stability (DARTS) accelerated small molecules target discovery: Principles and application

Ying-Shan Ren et al.

Summary: DARTS is a novel target discovery approach that is particularly adept at screening small molecule targets without requiring any structural modifications. It can reveal drug-target interactions from cells or tissues and has been applied to uncover drug-action mechanisms.

Discovery of 5-{4-[(7-Ethyl-6-oxo-5,6-dihydro-1,5-naphthyridin-3-yl)methyl]piperazin-1-yl}-N-methylpyridine-2-carboxamide (AZD5305): A PARP1-DNA Trapper with High Selectivity for PARP1 over PARP2 and Other PARPs

Jeffrey W. Johannes et al.

Summary: PARP inhibitors have obtained regulatory approval in oncology for tumors with homologous recombination repair deficiency, including those with BRCA mutations. However, some inhibitors have failed in combination with first-line chemotherapies due to overlapping hematological toxicities. Current PARP inhibitors lack selectivity for PARP1, which may contribute to toxicity.

Monocarboxylate transporter-1 (MCT1) protein expression in head and neck cancer affects clinical outcome

Martin Leu et al.

Summary: The study evaluated the potential of MCT1 and MCT4 as biomarkers in HNSCC patients, finding that positive staining for MCT1 was strongly associated with worsened PFS and OS, indicating it could be a promising biomarker in HNSCC treated by primary RCT.

High expression of monocarboxylate transporter 4 (MCT 4), but not MCT 1, predicts poor prognosis in patients with non-small cell lung cancer

Ying-Hui Tong et al.

Summary: In this study, high expression of MCT4 was found to be associated with worse overall and progression-free survival in non-small cell lung cancer patients and identified as an independent predictive indicator. Advanced cancer stage and high MCT4 level were also determined as independent predictive indicators for poor outcomes. No significant differences were observed in subgroup and interaction analyses in different clinical characteristic groups.

A Comparison of Two Stability Proteomics Methods for Drug Target Identification in OnePot 2D Format

Yingrong Xu et al.

Summary: Stability proteomics techniques have emerged as an attractive alternative in drug target engagement studies, with SPROX and TPP being representative methods. The adoption of the OnePot strategy has increased throughput and efficiency, with the OnePot 2D format offering improved analysis specificity. However, there is still a lack of systematic evaluation of the OnePot 2D format and a comparison between SPROX and TPP.

Discovery of 5-{2-[5-Chloro-2-(5-ethoxyquinoline-8-sulfonamido)phenyl]ethynyl}-4-methoxypyridine-2-carboxylic Acid, a Highly Selective in Vivo Useable Chemical Probe to Dissect MCT4 Biology

Timo Heinrich et al.

Summary: This study describes the discovery and optimization of a novel class of MCT4 inhibitors that inhibit lactate efflux and reduce cellular viability in MCT4 high expressing cells. The optimized compound 18n showed direct target interaction with MCT4 and allowed assessment of lactate modulation and antitumor activity in a mouse tumor model. Compound 18n represents a valuable tool for investigating selective MCT4 inhibition and its effect on tumor biology.
