Targeted oral peptide delivery using multi-unit particulates: Drug and permeation enhancer layering approach

Summary: This work employed layering techniques in a fluidized bed dryer to create a configuration for oral delivery of peptides, which involved coating active peptides, permeation enhancers, and polymers to control release. Formulations were designed to disintegrate at different pH values and trigger release at specific sites in the gastrointestinal tract, with dissolution studies confirming the duration of release. Pharmacokinetic studies in beagle dogs showed that a high disintegration pH was beneficial in enhancing bioavailability.

Polyphosphate coatings: A promising strategy to overcome the polycation dilemma

Summary: This study aimed to develop a drug delivery system that changes the zeta potential by decorating lipid-based nanocarriers with a polycationic cell penetrating peptide (CPP) and masking these cationic substructures with polyphosphates. The polyphosphate-coated PLL-decorated nanocarriers showed improved cellular uptake and significant conversion of zeta potential.
