Novel Small-Molecule Troponin Activator Increases Cardiac Contractile Function Without Negative Impact on Energetics

Huamei He et al.

Summary: The study found that the novel myotrope TA1 can increase myocardial contractility without impairing diastolic function or depleting the cardiac energy reserve, making it a potentially superior alternative to traditional inotropes in heart failure management.

Making waves: A proposed new role for myosin-binding protein C in regulating oscillatory contractions in vertebrate striated muscle

Samantha P. Harris

Summary: Myosin-binding protein C (MyBP-C) plays a crucial role in regulating muscle performance by stabilizing a newly discovered super-relaxed state of myosin on thick filaments and prolonging the on state of actin on thin filaments. Its dual interactions with both myosin and actin are important for understanding its new functions and implications in muscle contraction and relaxation. Further exploration of its molecular mechanisms as a wave maker or a wave breaker will provide insights into previously unrecognized regulatory mechanisms in vertebrate striated muscle.

Cardiac Myosin Activation with Omecamtiv Mecarbil in Systolic Heart Failure

John R. Teerlink et al.

Summary: Among patients with heart failure and reduced ejection fraction, those who received omecamtiv mecarbil had a lower incidence of heart-failure events or cardiovascular death compared to those who received placebo over a median of 22 months.

Omecamtiv mecarbil evokes diastolic dysfunction and leads to periodic electromechanical alternans

Gabor A. Fulop et al.

Summary: Omecamtiv mecarbil (OM) is a promising novel drug for improving cardiac contractility, but may have previously unrecognized side effects. Concentration-dependent improvements in cardiac function were observed with OM, although higher concentrations were associated with hypotension and diastolic dysfunction.

Evidence-Based Assessment of Genes in Dilated Cardiomyopathy

Elizabeth Jordan et al.

Summary: A systematic curation of 51 genes related to DCM revealed that 19 genes have high evidence (12 definitive/strong, 7 moderate), but these genes only explain a minority of cases, leaving the remainder of DCM genetic architecture incompletely addressed.

Amino terminus of cardiac myosin binding protein-C regulates cardiac contractility

Thomas L. Lynch et al.

Summary: Phosphorylation and dephosphorylation of cardiac myosin binding protein-C play crucial roles in regulating cardiac contraction and actomyosin interactions. The amino terminus region of cMyBP-C is essential for maintaining normal cardiac structure and function, with its deletion resulting in dilated cardiomyopathy.
