Lipid21: Complex Lipid Membrane Simulations with AMBER

Callum J. Dickson et al.

Summary: This study extends the modular AMBER lipid force field to include anionic lipids, PUFA lipids, and sphingomyelin, enabling the simulation of realistic cell membrane lipid compositions, including raft-like domains. The revised torsion parameters of the head groups and updated hydrocarbon chain parameters improve the accuracy of the force field. Extensive validation runs show good agreement with experimental measurements.

Drug-Membrane Interactions: Effects of Virus-Specific RNA-Dependent RNA Polymerase Inhibitors Remdesivir and Favipiravir on the Structure of Lipid Bilayers

Markus Fischer et al.

Summary: The translation introduces the importance of remdesivir and favipiravir, two RNA-dependent RNA polymerase inhibitors, in the treatment of COVID-19, as well as their impact on lipid bilayer membrane structures. While remdesivir disrupts membrane structure, it only slightly affects erythrocyte membrane integrity, while favipiravir shows no indications of affecting membrane structure in experiments.

An in vitro study on anti-carcinogenic effect of remdesivir in human ovarian cancer cells via generation of reactive oxygen species

Chang Min Lee et al.

Summary: This study demonstrates that remdesivir has an anti-carcinogenic effect on human ovarian cancer cell line SKOV3 cells. Remdesivir reduces cell viability, induces apoptosis, and affects the expression of important proteins involved in cell survival and apoptosis. These effects may be mediated through the up-regulation of reactive oxygen species.

Interaction of Remdesivir with Clinically Relevant Hepatic Drug Uptake Transporters

Anne T. Nies et al.

Summary: Remdesivir has been approved for COVID-19 treatment, but the study indicates that OATP1B1 and its genetic variants, OATP1B3, OATP2B1 and OCT1, are not relevant for the hepatic uptake of the drug in humans. This suggests that there are no clinically relevant transporter-mediated drug interactions expected due to the rapid clearance of remdesivir.

Favipiravir for the treatment of patients with COVID-19: a systematic review and meta-analysis

Toshie Manabe et al.

Summary: The study suggests that favipiravir shows promising efficacy in treating patients with COVID-19, inducing viral clearance within 7 days and contributing to clinical improvement within 14 days. The drug is especially effective for patients with mild-to-moderate illness. However, additional well-designed studies are needed to determine the optimal dosage and duration of treatment.

Extracorporeal membrane oxygenation may decrease the plasma concentration of remdesivir in a patient with severe coronavirus disease 2019

Satoshi Ide et al.

Summary: Remdesivir is an antiviral drug that has shown clinical improvement and accelerated recovery in patients, but there is a lack of pharmacokinetic analysis in patients with severe COVID-19 requiring ECMO treatment. Monitoring plasma concentrations of remdesivir and GS-441524 in ECMO patients may be necessary to ensure proper dosing and effectiveness.

Phenolic compounds alter the ion permeability of phospholipid bilayers via specific lipid interactions

Sheikh Hossain et al.

Summary: This study combines experimental and molecular dynamics simulation methods to investigate the interactions between six phenolic compounds and membranes. The results show that specific phenolic-lipid interactions play a critical role in altering ion permeability, independent of lipophilicity.
