Abietic, maleopimaric and quinopimaric dipeptide Ugi-4CR derivatives and their potency against influenza A and SARS-CoV-2

Summary: A series of abietane diterpene derivatives have been synthesized by the Ugi-four component reaction (Ugi-4CR), and their activity has been tested. Five dipeptide derivatives showed good selectivity and low IC50 values against influenza virus, with compound 11 exhibiting the best activity. Additionally, one of the compounds showed weak activity against SARS-CoV-2. Therefore, diterpene acids-Ugi products have the potential to be new effective anti-influenza compounds.

Comparison of structural properties of cyclosporin A and its analogue alisporivir and their effects on mitochondrial bioenergetics and membrane behavior

Summary: This paper investigates the effects of the MPT pore inhibitor cyclosporin A (CsA) and its non-immunosuppressive analogue alisporivir (Ali) on rat skeletal muscle mitochondria. The results showed that both CsA and Ali can increase the calcium capacity of the mitochondria at a standard concentration, without affecting oxidative phosphorylation. However, at higher concentrations, both agents suppress oxygen consumption by the mitochondria, with Ali showing a stronger effect. This suppression is accompanied by a decrease in the membrane potential and is likely due to the inhibition of electron transport along the mitochondrial respiratory chain. Both CsA and Ali also decrease the overall fluidity of liposomes and mitochondrial membranes, suggesting that their lipophilic nature and ability to accumulate in lipid membranes may contribute to the decrease in electron transport efficiency and bioenergetics of mitochondria.
