EMSY inhibits homologous recombination repair and the interferon response, promoting lung cancer immune evasion

Antonio Marzio et al.

Summary: NSCLCs with KEAP1 mutations show resistance to immunotherapy due to EMSY accumulation suppressing immune response. KEAP1 loss results in stabilized EMSY, causing HRR defects and sensitivity to PARP inhibitors. The accumulation of EMSY impairs immune signaling and promotes immune evasion. Activation of the type I interferon response using a STING agonist inhibits KEAP1-mutant tumor growth, suggesting a potential therapeutic strategy for NSCLC patients with KEAP1 alterations.

Weed Suppression and Molecular Mechanisms of Isochlorogenic Acid A Isolated from Artemisia argyi Extract via an Activity-Guided Method

Le Chen et al.

Summary: In this study, the allelopathic active constituents of Artemisia argyi were identified using activity-guided isolation and UPLC-QTOF-MS. The results showed that phenolic compounds are the main allelochemicals in A. argyi. Additionally, a novel allelochemical, ICGAA, with broad herbicidal activity was discovered. Further analysis revealed that ICGAA inhibits weed growth by inhibiting the diterpenoid biosynthesis pathway, especially gibberellins (GAs). This study provides new material sources and theoretical support for the development of biological herbicides in agroecosystems.

NINJ1 mediates plasma membrane rupture during lytic cell death

Nobuhiko Kayagaki et al.

Summary: Plasma membrane rupture is a cataclysmic event in lytic cell death, releasing damage-associated molecular patterns to propagate the inflammatory response. NINJ1 protein plays an essential role in inducing PMR, with Ninj1-deficient macrophages showing impaired release of intracellular proteins and distinctive ballooned morphology. This study overturns the long-held belief that cell death-related PMR is a passive event, highlighting the active role of NINJ1 in mediating PMR.

Evaluation of inhibitory effects of geniposide on a tumor model of human breast cancer based on 3D printed Cs/Gel hybrid scaffold

Kena Lv et al.

Summary: Traditional Chinese medicine therapy, specifically geniposide, has shown to have anti-cancer effects with no obvious side effects on the human body. The use of 3D printed Cs/Gel composite scaffolds provided a more accurate platform for drug screening, demonstrating the effectiveness of geniposide in inhibiting proliferation and promoting apoptosis of breast cancer cells (MCF-7) in a time- and dose-dependent manner. Live/Dead staining, Hematoxylin-Eosin (H&E) staining and SEM evaluation further confirmed the anti-cancer effects of geniposide.

Dendritic Cell-Based Immunotherapy in Lung Cancer

Dieter Stevens et al.

Summary: Immune therapy plays a significant role in the treatment of lung cancer patients, with potential for further exploration. Combining immune checkpoint blockade with other strategies may be a way to fully harness the immune system's anti-tumor potential. Dendritic cells, with their unique ability to initiate and regulate T cell responses, show promise as tools for immunotherapy in lung cancer.
