Repurposing α-Adrenoreceptor Blockers as Promising Anti-Virulence Agents in Gram-Negative Bacteria

Summary: Antimicrobial resistance is a pressing global public health issue. This study investigates the potential of blocking adrenergic receptors as a strategy to reduce bacterial virulence and inhibit the development of resistance. The researchers found that terazosin, an alpha-adrenergic blocker, showed promising anti-quorum sensing (QS) and anti-virulence activities against two bacteria strains. It effectively reduced pigment production in one strain and inhibited biofilm formation and down-regulated QS genes in another strain.

Summary: The study finds that the alpha-adrenergic antagonist terazosin has an anti-virulence effect on S. Typhimurium. Terazosin reduces the adhesion, biofilm formation, invasion, and intracellular replication of S. Typhimurium. It also interferes with quorum sensing and bacterial espionage, as well as down-regulates the expression of the type three secretion system (T3SS) encoding genes.

Repurposing of antidiabetics as Serratia marcescens virulence inhibitors

Summary: Vildagliptin and metformin reduced the expression of virulence encoding genes in S. marcescens, but did not show anti-virulence activities in vitro or in vivo. On the other hand, sitagliptin significantly inhibited virulence factors in vitro, reduced their expression, and protected mice from S. marcescens infection. Docking study revealed that sitagliptin was more active than metformin and fully bound to the SmaR receptor, suggesting potential as a therapeutic target.
