Multi-stage structure-based virtual screening approach towards identification of potential SARS-CoV-2 NSP13 helicase inhibitors

Summary: In this study, a multi-stage structure-based drug discovery approach was used to identify a potential inhibitor targeting SARS-COV-2 NSP13 helicase enzyme. A 3D pharmacophore was generated based on protein-ligand interaction fingerprint (PLIF) study, and the ZINC database was screened to retrieve potential hits. The best hit was further analyzed using molecular dynamics simulations and binding free energy calculations, confirming its potential as a potent NSP13 helicase inhibitor.

Compound 12a showed the strongest inhibitory effect on various cancer cell lines and exhibited high affinity for VEGFR-2. Furthermore, all tested compounds demonstrated high safety profiles in non-cancerous cell lines.

2-Pyridone natural products as inhibitors of SARS-CoV-2 main protease

These 2-pyridone compounds were also evaluated for their drug-like properties, indicating the potential for rapid discovery of a COVID-19 treatment.
