Synthesis, Characterization and Biodistribution of GdF3:Tb3+@RB Nanocomposites

Summary: In this study, a nanocomposite for X-ray-induced photodynamic therapy and computed tomography was developed using PEG-capped GdF3:Tb3+ scintillating nanoparticles conjugated with Rose Bengal. The synthesized nanoparticles had an elongated spindle-like clustered morphology with an orthorhombic structure, and the presence of PEG coating and Rose Bengal conjugates was confirmed. Fluorescent resonant energy transfer between scintillating nanoparticles and Rose Bengal was efficiently detected upon X-ray irradiation of the nanocomposite solution.

This work provides a new strategy for deep-tissue PDT and immune response triggered by soft X-ray, overcoming the depth barriers faced by traditional photoactivated PDT or ICD with visible and near infrared light.

Activatable Second Near-Infrared Fluorescent Probes: A New Accurate Diagnosis Strategy for Diseases

Challenges and perspectives of activatable NIR-II fluorescent probes are also discussed in this review.

The multifunctionality of this method allows for covalent incorporation of Rose Bengal into nanostructures and enhancement of internalization by tumor cells through DNA aptamer modification. The aptamer-labeled nanoparticles prepared for PDT can potentially be used for clinical cancer treatment.
