The molecular pathogenesis of repeat expansion diseases

Yuzo Fujino et al.

Summary: In summary, short tandem repeat expansions in the genome are associated with various monogenic diseases, especially neurological disorders. These diseases can be categorized into coding repeat expansions that cause polypeptide toxicity, and non-coding repeat expansions that lead to RNA toxicity. Research is currently focused on the pathological mechanisms involving repeat RNAs and repeat polypeptides, as well as potential therapeutic strategies against these diseases.

Facultative heterochromatin formation in rDNA is essential for cell survival during nutritional starvation

Hayato Hirai et al.

Summary: During nutrient starvation, fission yeast cells assemble facultative heterochromatin in rDNA, which leads to transcriptional repression. Histone modifications and histone deacetylases play important roles in this process.

Low Complexity Induces Structure in Protein Regions Predicted as Intrinsically Disordered

Mariane Goncalves-Kulik et al.

Summary: This study provides evidence that low complexity regions (LCRs) within intrinsically disordered regions (IDRs) of proteins can induce local structure. By predicting IDRs in the human proteome and analyzing structures in the Protein Data Bank (PDB), the researchers found that LCRs were more likely to have structural information assigned to them compared to surrounding IDRs. The most frequent LCRs containing E (Glu) or G (Gly) were found to induce helical or coil structures. These findings suggest a structuring role of LCRs within IDRs.

FBXO44 promotes DNA replication-coupled repetitive element silencing in cancer cells

Jia Z. Shen et al.

Summary: The study identified FBXO44 as an essential repressor of repetitive elements in cancer cells, with its inhibition leading to DNA replication stress and viral mimicry. This promotes decreased tumorigenicity, increased immunogenicity, and enhanced immunotherapy response in cancer cells.

Between Interactions and Aggregates: The PolyQ Balance

Pablo Mier et al.

Summary: Polyglutamine (polyQ) regions are consecutive runs of glutamine residues, which are associated with various diseases. Studies suggest that polyQ may serve as a motif for modulation of protein-protein interactions. However, long polyQ regions could lead to an excess of interactions and the formation of aggregates.

Summary: ATAD2 functions as a co-activator in DNA replication and histone recognition, and is up-regulated in various forms of cancer, correlating with poor prognosis. The study reveals the recognition of histone acetyllysine modifications by ATAD2 bromodomain, illustrating specific interactions that make it a potential target for cancer therapeutics.

Between Interactions and Aggregates: The PolyQ Balance

Pablo Mier et al.

Summary: Polyglutamine (polyQ) regions are consecutive runs of glutamine residues, mainly associated with polyQ-related diseases. Besides causing protein aggregation diseases, polyQ may also serve as a motif for modulating protein-protein interactions. Long polyQ regions may lead to an excess of interactions and aggregate formation.
