Development of off-the-shelf hematopoietic stem cell-engineered invariant natural killer T cells for COVID-19 therapeutic intervention

Yan-Ruide Li et al.

Summary: Researchers have successfully developed a method to produce therapeutic levels of (HSC)-H-Allo-iNKT cells, which closely resemble endogenous human iNKT cells in phenotypes and functionalities. These cells are capable of directly killing SARS-CoV-2 infected cells and suppressing inflammation caused by SARS-CoV-2 infection in cell culture. Additionally, these cells pose no risk of GvHD and are resistant to T cell-mediated allorejection.

Targeting Immunosuppressive Tumor-Associated Macrophages Using Innate T Cells for Enhanced Antitumor Reactivity

Yan-Ruide Li et al.

Summary: This study evaluates the potential of innate T cells for antitumor therapy, targeting both tumor cells and immunosuppressive tumor-associated macrophages. Recent developments in innate immune cell CAR therapy have opened up new possibilities for overcoming challenges in traditional therapies, especially in the realm of tumor treatment and immunotherapy.

Development of Stem Cell-Derived Immune Cells for Off-the-Shelf Cancer Immunotherapies

Yan-Ruide Li et al.

Summary: Cell-based cancer immunotherapy has revolutionized the treatment of hematological malignancies, with CAR-T cell therapies achieving unprecedented clinical response rates. The autologous nature of current CAR-T cell products poses challenges, while allogeneic cell products face concerns such as GvHD and allorejection. Advances in developing stem cell-engineered allogeneic cell therapies aim to overcome these limitations.

GVHD Prophylaxis 2020

Mahasweta Gooptu et al.

Summary: GVHD, both acute and chronic, is a major complication of allogeneic transplantation, and its prevention is crucial for the success of transplants. Current standards revolve around calcineurin inhibitors, but promising translational advances targeting T-cell regulation and co-stimulatory pathways are emerging. These approaches are based on a pathophysiology perspective and show potential for transforming GVHD prophylaxis in the near future.

Development of allogeneic HSC-engineered iNKT cells for off-the-shelf cancer immunotherapy

Yan-Ruid Li et al.

Summary: Through hematopoietic stem cell gene engineering and in vitro differentiation, human allogeneic iNKT cells can be produced at high yield and purity, effectively targeting tumor cells with low immunogenicity. These cells can be further engineered to enhance tumor targeting or reduce immunogenicity for potential cancer therapy.
