Outgrowth Endothelial Cell Conditioned Medium Negates TNF-α-Evoked Cerebral Barrier Damage: A Reverse Translational Research to Explore Mechanisms

Summary: Improved understanding of the mechanisms underlying cerebral ischemic injury is crucial for developing effective therapeutics for stroke. This study found significant elevations in TNF-alpha levels in plasma samples from ischemic stroke patients at different time points after stroke. The inflammatory cytokine was found to impair blood-brain barrier integrity and function, leading to early death. However, co-treatment with outgrowth endothelial cell-derived conditioned media (OEC-CM) mitigated the detrimental effects of TNF-alpha on the blood-brain barrier. OEC-CM showed vasculoprotective effects by suppressing anti-angiogenic factors, reducing oxidative stress and apoptosis, and improving the adhesive and tubulogenic properties of brain microvascular endothelial cells and OECs.
