Candidates for Repurposing as Anti-Virulence Agents Based on the Structural Profile Analysis of Microbial Collagenase Inhibitors

Summary: Pharmacological inhibition of bacterial collagenases (BC) is an effective strategy to block bacterial virulence without inducing drug resistance. In this study, the chemical structures of Clostridium perfringens collagenase A (ColA) inhibitors were analyzed and various scoring methods were used to predict new compounds with ColA inhibitory activity. The results showed that compounds containing at least one aromatic ring, particularly a nitrobenzene fragment, exhibit higher potency as BC inhibitors. Furthermore, molecular descriptors were identified as filters to identify potential BC inhibitors, and the DrugBank database was screened to identify 100 potential BC inhibitors, including 10 approved drugs. Benzthiazide, entacapone, and lodoxamide were selected as the most promising candidates for in vitro testing based on their pharmacotoxicological profiles.

The Polyphenolic Profile and Antioxidant Activity of Five Vegetal Extracts with Hepatoprotective Potential

Summary: Oxidative stress is a major factor in human functional disorders, and this study aimed to obtain standardized plant extracts containing active compounds that protect liver cells against free radicals. The extracts were analyzed to identify and quantify therapeutic compounds using UHPLC-HRMS/MS. The extracts contained various flavonoids, phenolic acids, and polyphenols. In vitro experiments confirmed the extracts' antioxidant activity, and a correlation was found between the content of polyphenolic compounds and the antioxidant effect. The extracts also showed hepatoprotective potential in silico. This study validates the use of these extracts in phytotherapy due to their ability to reduce oxidative stress and protect liver cells.
