CircCEMIP promotes anoikis-resistance by enhancing protective autophagy in prostate cancer cells

Ying Yu et al.

Summary: This study demonstrates that circ_0004585 plays an oncogenic role in the invasion and metastasis of prostate cancer by targeting the miR-1248/TM9SF4 axis and promoting anoikis resistance. These findings provide new insights into therapeutic strategy development for metastatic prostate cancer.

Identification and Validation of a Gene Signature for Lower-Grade Gliomas Based on Pyroptosis-Related Genes to Predict Survival and Response to Immune Checkpoint Inhibitors

Guichuan Lai et al.

Summary: Pyroptosis-related gene signature has a significant impact on the prognosis and efficacy of immune checkpoint inhibitors (ICIs) in lower-grade gliomas (LGGs). High-risk patients have worse survival outcomes and distinct differences in tumor immune microenvironment, mutation status, and response to therapy compared to low-risk patients. These findings are valuable in predicting prognosis and guiding ICIs treatment for LGG patients.

A Novel Classification Model for Lower-Grade Glioma Patients Based on Pyroptosis-Related Genes

Yusheng Shen et al.

Summary: This study identified two pyroptosis phenotypes in lower-grade glioma, which exhibited differences in cell infiltration characteristics and clinical characteristics. The PyroScore model was constructed to measure the level of pyroptosis in each patient, allowing for stratified prognosis and personalized management.

Tumor cell-derived ANGPTL2 promotes β-catenin-driven intestinal tumorigenesis

Haruki Horiguchi et al.

Summary: Tumor cell-derived ANGPTL2 promotes beta-catenin-driven intestinal tumorigenesis by upregulating OB-cadherin expression and blocking destruction complex-independent proteasomal degradation of beta-catenin proteins. It also leads to decreased cell surface integrin alpha 5 beta 1 expression.

HuR-dependent SOD2 protein synthesis is an early adaptation to anchorage-independence

Yeon Soo Kim et al.

Summary: During metastasis, cancer cells need to adapt to loss of anchorage and resistance to anoikis. One important adaptation is the increase in antioxidant capacity and restoration of cellular redox balance. In ovarian cancer cells, an early response to detachment, a critical step in metastasis associated with increased oxidative stress, is the increase in mitochondrial superoxide dismutase SOD2 protein expression. This is achieved through the translational regulatory mechanism involving the cytosolic accumulation of the RNA binding protein HuR/ELAVL1 and the activation of mitogen-activated kinase p38.

Inflammation: the incubator of the tumor microenvironment

Dominic Denk et al.

Summary: Inflammatory microenvironment is a prerequisite and fuel for almost all cancers. The complex interplay of different cell types in the tumor microenvironment determines patient outcome. In addition to tumor cells, local and recruited nonmalignant cells as well as the intestinal microbiome actively promote cancer progression in the tumor microenvironment.

A Novel Six Autophagy-Related Genes Signature Associated With Outcomes and Immune Microenvironment in Lower-Grade Glioma

Tao Lin et al.

Summary: A powerful predictive signature related to autophagy was successfully constructed for prognosis prediction in patients with Lower-grade gliomas, which was identified as an independent risk predictor of overall survival. Functional enrichment analysis revealed that the autophagy-related signatures were mainly involved in phagosome and immune-related pathways. Significant differences in immune cell infiltration and immunotherapy targets were found between risk groups, indicating potential implications for personalized immunotherapy in LGG.

Angiopoietin-like protein 2 decreases peritoneal metastasis of ovarian cancer cells by suppressing anoikis resistance

Yuko Takeshita et al.

Summary: The study indicates that ANGPTL2 expression in ovarian cancer cells represses peritoneal metastasis by suppressing anoikis resistance. Cells expressing ANGPTL2 showed decreased formation of metastasis in the mouse intraperitoneal xenograft model. This suggests a potential therapeutic target for inhibiting ovarian cancer peritoneal metastasis.

TISCH: a comprehensive web resource enabling interactive single-cell transcriptome visualization of tumor microenvironment

Dongqing Sun et al.

Summary: Cancer immunotherapy targeting co-inhibitory pathways by checkpoint blockade has shown significant efficacy, but only a minority of patients respond due to tumor microenvironment heterogeneity. TISCH is a large curated database that integrates single-cell transcriptomic profiles from 76 high-quality tumor datasets across 27 cancer types, providing a user-friendly interface for exploring gene expression atlas in the TME from multiple cancer types.
