Antifungal and Anti-Virulent Activity of Origanum majorana L. Essential Oil on Candida albicans and In Vivo Toxicity in the Galleria mellonella Larval Model

Summary: This study investigated and compared the antifungal activity and essential oil composition of naturally growing and cultivated samples of marjoram. The results showed that both samples of marjoram essential oil exhibited strong antifungal activity against Candida spp. Additionally, the essential oils showed inhibition of biofilm formation and germ-tube formation, which are virulence factors of Candida albicans.

This review article focuses on the efforts to find effective drugs against azole-resistant candidiasis, covering azole-based compounds, dual target drugs, and new scaffolds. The mechanism of action and SAR studies are also discussed. Experts believe that novel scaffolds/compounds can be found using methods like virtual screening and fragment-based drug discovery.

Design, synthesis and biological evaluation of imidazole and triazole-based carbamates as novel aromatase inhibitors

Summary: Novel aromatase inhibitors based on triazole and imidazole-based carbamate derivatives were designed and synthesized, with compounds 13a and 15c showing dose-dependent inhibition of cell viability on the human breast cancer cell line MCF7. Docking simulations were also carried out to elucidate the binding modes of these active compounds to target human aromatase at a molecular level.

Inhibitory effect of polyunsaturated fatty acids alone or in combination with fluconazole on Candida krusei biofilms in vitro and in Caenorhabditis elegans

Summary: The incidence of infections caused by non-albicans Candida species, including Candida krusei, is increasing, and these infections show resistance to fluconazole and can form biofilms. This study found that linoleic acid (LA) and gamma-linolenic acid (GLA) can inhibit C. krusei biofilms and increase their susceptibility to fluconazole by inducing oxidative stress, cell membrane damage, and disrupting efflux pump activity. These PUFAs also prolonged the lifespan of infected nematodes and showed a synergistic effect with fluconazole, suggesting a potential new treatment strategy for combating infections caused by C. krusei.

However, there are gaps in knowledge concerning specific mechanisms underlying G. mellonella-fungal interactions that need to be addressed in order to strengthen the model's viability for studying fungal virulence in the future.
