Dietary Interventions in Autosomal Dominant Polycystic Kidney Disease

Lauren Pickel et al.

Summary: Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive growth of renal cysts and loss of functional nephrons. Evidence suggests that dietary interventions such as caloric restriction, intermittent fasting, and ketogenic diets have the potential to slow disease progression and confer metabolic benefits.

Metformin Therapy in Autosomal Dominant Polycystic Kidney Disease: A Feasibility Study

Godela M. Brosnahan et al.

Metformin improves relevant disease parameters in an autosomal dominant polycystic kidney disease mouse model

Nuria M. Pastor-Soler et al.

Summary: This study tested the effectiveness of metformin as a therapy for ADPKD in a relevant mouse model and found that metformin reduced kidney cysts, improved kidney function, lowered blood pressure and urea nitrogen levels, and reduced inflammation and injury markers. These findings have implications for new ADPKD therapies in patients.

Negative regulation of AMPK signaling by high glucose via E3 ubiquitin ligase MG53

Peng Jiang et al.

Summary: High glucose suppresses AMPK signaling through MG53-mediated AMPK alpha degradation and deactivation, as well as ROS-dependent inhibition of AMPK alpha phosphorylation at T172. These findings elucidate the mechanism of impaired AMPK signaling in overnutrition-related diseases and underscore the importance of maintaining the yin-yang balance of AMPK signaling for metabolic homeostasis.
