Posthemorrhagic hydrocephalus associates with elevated inflammation and CSF hypersecretion via activation of choroidal transporters

Sara Diana Lolansen et al.

Summary: This study aimed to elucidate the molecular coupling between hemorrhagic events and the development of posthemorrhagic hydrocephalus (PHH), as well as the inflammatory profile of PHH pathogenesis. The findings suggest that excessive cerebrospinal fluid (CSF) secretion may contribute to PHH development, likely due to hyperactivity of choroid plexus transporters. Hemorrhage-induced inflammation detected in CSF and choroid plexus tissue may represent the underlying pathology of PHH.

Transcriptional profiling of transport mechanisms and regulatory pathways in rat choroid plexus

Soren N. Andreassen et al.

Summary: This study provides important molecular tools for studying pathologies related to disturbed brain fluid homeostasis by analyzing the transcriptome of the choroid plexus in rats. The results show similarities in transport protein transcript abundance between species and provide a list of potential transport mechanisms and regulatory candidates involved in cerebrospinal fluid secretion.

Elevated CSF inflammatory markers in patients with idiopathic normal pressure hydrocephalus do not promote NKCC1 hyperactivity in rat choroid plexus

Sara Diana Lolansen et al.

Summary: The study found elevated levels of certain inflammatory markers in the CSF of iNPH patients, but these markers did not affect the activity of choroidal NKCC1 in experiments. This indicates that the molecular mechanisms underlying ventriculomegaly in iNPH and its potential connection to inflammation still need to be elucidated.

Molecular mechanisms of brain water transport

Nanna MacAulay

Summary: Our brains consist of 80% water, which is continuously shifted between different compartments and cell types during physiological and pathophysiological processes. Disturbances in brain water homeostasis occur with pathologies such as brain oedema and hydrocephalus. Although aquaporins greatly accelerate the movement of water, they do not suffice to explain overall brain water movement, suggesting the presence of unknown molecular mechanisms.

CSF Secretion Is Not Altered by NKCC1 Nor TRPV4 Antagonism in Healthy Rats

Steven W. Bothwell et al.

Summary: Acute antagonism of NKCC1 and TRPV4 proteins at the choroid plexus does not reduce cerebrospinal fluid (CSF) secretion in healthy rats. Only the positive control acetazolamide significantly decreased CSF secretion rate.

TRPV4 channel activation induces the transition of venous and arterial endothelial cells toward a pro-inflammatory phenotype

Kathia Beddek et al.

Summary: Activation of TRPV4 channels in endothelial cells can lead to increased intracellular Ca2+ levels, actin cytoskeleton rearrangement, decreased barrier permeability, increased adhesion molecule expression, and induction of apoptosis, suggesting a role in transitioning endothelial cells towards a pro-inflammatory phenotype.
