Summary: This study reveals that Bcl-2 inhibitors mediate cell cycle arrest at the G0/G1 phase, rather than their proapoptotic activity, to play a critical role in blocking mammarenavirus multiplication. Additionally, the Bcl-2 inhibitor ABT-737 exhibited moderate efficacy against mammarenaviruses in vivo, showcasing broad-spectrum antiviral activities against various species and SARS-CoV-2. These results suggest that Bcl-2 inhibitors might be repositioned as effective broad-spectrum antivirals.
