Halogenation Activity of Mammalian Heme Peroxidases

Summary: This article reviews the unique characteristics and mechanisms of mammalian heme peroxidases in oxidizing (pseudo)halides under physiological conditions. It highlights the importance of thyroid peroxidase and peroxidasin in thyroid hormone synthesis and basement membrane formation, as well as the properties and (pseudo)halogenated products of immunologically relevant proteins. The potential roles of these (pseudo)halogenated products are further discussed.

Fluorescent Probes for HOCl Detection in Living Cells

Summary: Hypochlorous acid (HOCl) plays a crucial role in the immune system but its high reactivity can also lead to the development of diseases. Therefore, developing highly sensitive and selective HOCl biosensors is important. This review summarizes the achievements in the development of chemosensors for HOCl detection in biological systems from 2016 to 2021. The challenges in designing and applying small-molecule probes are discussed, and recommendations are given for their future application.

MPO binds to extracellular biomolecules, leading to localized damage on specific sites and species, affecting cellular dysfunction and altered gene expression. MPO's interaction with damaged ECM components may explain its accumulation at sites of inflammation.
