A Programmable, DNA-Exclusively-Guided Argonaute DNase and Its Higher Cleavage Specificity Achieved by 5′-Hydroxylated Guide

Characterization of a Programmable Argonaute Nuclease from the Mesophilic Bacterium Rummeliibacillus suwonensis

Summary: This study discovered three active prokaryotic Argonautes (pAgos) with about 50% sequence identity to KmAgo, which have typical DNA-guided DNA target cleavage ability. Among them, RsuAgo from Rummeliibacillus suwonensis was mainly explored for its ability to cleave both DNA and RNA targets, especially with short guide DNAs in the range of 16-20 nt. RsuAgo-mediated RNA target cleavage showed a preference for a guide strand with a 5'-terminal A residue. Furthermore, RsuAgo was found to be able to cleave double-stranded DNA in a low-salt buffer at 37 degrees C, providing a new tool for DNA and RNA manipulation at moderate temperatures.

Summary: This study reveals a short pAgo protein in the archaeon Sulfolobus islandicus, along with its genetically associated proteins Aga1 and Aga2, provide robust anti-viral protection through abortive infection. Aga2 is a toxic transmembrane effector that binds anionic phospholipids, resulting in membrane depolarization and cell killing. Ago and Aga1 form a stable complex with nucleic acid-directed recognition ability and directly interact with Aga2, suggesting an immune sensing mechanism.

The combination of CbAgo and RecB(exo-)C represents the most efficient mesophilic DNA-guided DNA-cleaving programmable endonuclease for in vitro use.
