Summary: MANF is crucial for the survival and function of mouse beta-cells, but its role in human beta-cell development and function remains unclear. Lack of MANF in humans leads to diabetes due to increased ER stress and impaired beta-cell function. Knocking out MANF gene in human embryonic stem cells induces mild ER stress and impairs insulin-processing capacity of beta-cells both in vitro and in vivo.
