Summary: This study demonstrates a rational metabolic engineering approach to enhance the production of medium-chain-length polyhydroxyalkanoate (mcl-PHA) with enriched dominant monomer and improved material properties. Through systematic genetic engineering, the engineered strain showed significantly increased cell dry weight and mcl-PHA content in shake-flask culture. The thermophysical and mechanical properties of mcl-PHA were also improved with an enriched ratio of the dominant monomer.
