Measuring the density and viscosity of culture media for optimized computational fluid dynamics analysis of in vitro devices

Summary: In this study, the density and dynamic viscosity of culture media were measured and found to increase with the % volume of added fetal bovine serum. The viscosity of 5% FBS-supplemented RPMI-1640 was observed to significantly increase after 3 days of culture, with distinct differences between cell lines. These experimentally-derived values were applied in computational fluid dynamics analysis, showing differences in wall shear stress and pressure between fluid models.

Synergistic effects of fluid shear stress and adhesion morphology on the apoptosis and osteogenesis of mesenchymal stem cells

Summary: This study investigates the synergistic effect of adhesion morphology and fluid flow on the behavior of mesenchymal stem cells (MSCs). The results show that adhesion patterns with low circularity and large area are beneficial for osteogenic differentiation of MSCs. Additionally, fluid shear stress (FSS) promotes osteogenic differentiation and enhances nuclear transfer of the transcriptional regulator Yes-associated protein (YAP) in MSCs. These findings are important for the design of bio-functional substrates for cell transplantation in tissue engineering.

Probing Notch1-Dll4 signaling in regulating osteogenic differentiation of human mesenchymal stem cells using single cell nanobiosensor

Summary: This study established a LNA/DNA nanobiosensor for gene expression analysis in single hMSCs during osteogenic differentiation. The results showed that Notch1-Dll4 signaling plays an important role in regulating hMSCs osteogenic differentiation. Additionally, Dll4 was identified as a molecular signature of differentiated hMSCs during osteogenic induction, and the role of Notch1-Dll4 signaling in a 3D spheroid model was elucidated.
