Summary: Injectable hydrogels mimicking the natural extracellular matrix have advantages such as microinvasive implantation and complex defect repair, but current options for bone repair lack tissue adhesion, self-healing, and osteogenic activity. A new strategy involving mussel-inspired bisphosphonated injectable nanocomposite hydrogels with adhesive, self-healing, and osteogenic properties has been developed, showing excellent biocompatibility and promoting cell viability, proliferation, migration, and osteogenesis differentiation. The hydrogels, dually cross-linked, also demonstrate potential for bone regeneration with the successful healing of a rat cranial defect.

Summary: The use of hydrogels containing PTH and nHAP can promote cranial bone regeneration by enhancing the expression of osteogenic-related proteins and facilitating osteogenic differentiation of BMSCs through the Notch signaling pathway. Implantation of the hydrogel into a rat cranial defect model showed efficient bone regeneration compared to using the hydrogel alone or nHAP alone, indicating a promising therapeutic effect of combining nHAP and PTH for bone regeneration.
