2D-DIGE-MS Proteomics Approaches for Identification of Gelsolin and Peroxiredoxin 4 with Lymph Node Metastasis in Colorectal Cancer

Summary: A combination of fluorescence two-dimensional difference gel electrophoresis (2D-DIGE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry was used to identify potential markers for prognosis and intervention of colorectal cancer at different stages of lymph node metastasis (LNM). The study identified 18 differentially expressed proteins between non-LNM and LNM CRC tissues, including GSN and PRDX4, which were abundant in node-positive CRC. Downregulation of GSN and PRDX4 suppressed migration and invasiveness of CRC cells and induced G1/S cell cycle arrest. The EGFR/RhoA/PKC alpha/ERK pathways were found to play a critical role in the transcriptional activation of GSN and PRDX4 promoters.
