DNA methylation in adipocytes from visceral and subcutaneous adipose tissue influences insulin-signaling gene expression in obese individuals

Aneta Cierzniak et al.

Summary: By analyzing gene expression levels and DNA methylation in patients with obesity and insulin resistance, it was found that obese patients with insulin resistance have hypermethylation in gene promoters, with methylation rates negatively correlated with gene expression, suggesting excessive methylation as an epigenetic pathway linking obesity to subsequent insulin resistance.

Cholesterol-recognition motifs in the transmembrane domain of the tyrosine kinase receptor family: The case of TRKB

Cecilia Cannarozzo et al.

Summary: The study identified the presence of cholesterol-recognition amino acid consensus (CRAC) and its inverted version (CARC) in different species, with CARC only found in vertebrates. TRKB, a member of the RTK family, may play a crucial role in neuronal plasticity and exhibits a CARC motif in its transmembrane region. Molecular dynamics simulations showed that cholesterol interaction with the TRKB CARC motif mainly occurs at the central Y433 residue. The study suggests that CARC/CRAC motifs may be involved in the function of the RTK family transmembrane region.

PPARG Hypermethylation as the First Epigenetic Modification in Newly Onset Insulin Resistance in Human Adipocytes

Malgorzata Malodobra-Mazur et al.

Summary: Insulin resistance is a physiological state where insulin is unable to trigger glucose uptake properly. Epigenetic modifications, specifically DNA methylation, play a significant role in the pathogenesis of insulin resistance. Changes in DNA methylation of genes like PPARG appear to be crucial in the onset of insulin resistance.
