Atrial-specific hiPSC-derived cardiomyocytes in drug discovery and disease modeling

Mayel Gharanei et al.

Summary: The discovery and utilization of human-induced pluripotent stem cells (hiPSCs) have played a crucial role in investigating the pathophysiology of cardiovascular diseases. These cells can be generated from patients, edited on genome level, and differentiated into various cell types, providing a platform for regenerative medicine, disease modeling, drug testing, and tissue generation. Advancements have been made in differentiating patient-specific atrial cardiac myocytes (hiPSC-aCM) and atrial engineered heart tissue (aEHT), which have helped in understanding the mechanisms of atrial-specific diseases and evaluating the effectiveness of atrial-selective drugs. However, the immature phenotype of hiPSC-CMs compared to adult CMs remains a limitation.

Generation of heterozygous (MRli003-A-5) and homozygous (MRli003-A-6) voltage-sensing knock-in human iPSC lines by CRISPR/Cas9 editing of the AAVS1 locus

Fangfang Zhang et al.

Summary: Assessment of cardiomyocyte electrophysiological properties is crucial for understanding cardiac disorders and drug screening. Optical action potential imaging using a genetically encoded voltage-sensing fluorescent protein (VSFP) provides a high-throughput method for functional characterization of cardiomyocytes. This study successfully integrated VSFP into human induced pluripotent stem cells, creating two stable voltage indicator lines, which can be used for optical membrane potential recordings and have wide applications in cardiovascular biomedicine.

Sequential Defects in Cardiac Lineage Commitment and Maturation Cause Hypoplastic Left Heart Syndrome

Markus Krane et al.

Summary: The study revealed disruptions in gene programs and cellular processes in patients with hypoplastic left heart syndrome (HLHS), particularly in key aspects of fetal ventricular cardiogenesis such as the cell cycle and cardiomyocyte maturation. These abnormalities result in defects in early differentiation of cardiac progenitors, leading to left ventricular hypoplasia.

iPSC-derived cardiomyocytes from patients with myotonic dystrophy type 1 have abnormal ion channel functions and slower conduction velocities

Hugo Poulin et al.

Summary: The study revealed that cardiac issues in DM1 patients are mainly caused by ion channel abnormalities in cardiomyocytes, including Na+ channels and Ca2+ channels. These channel dysfunctions lead to prolonged action potential duration, slower velocities, and reduced conduction velocity.

Summary: Epidemiological studies have shown a genetic component to the risk of sudden cardiac death (SCD), especially in young individuals with rare, monogenic forms of heart disease. Despite comprehensive pathology and genetic evaluations, a proportion of SCD cases in young individuals remain unexplained, posing challenges in identifying family members at risk of SCD.
