Integrative metabolomics-genomics approach reveals key metabolic pathways and regulators of Alzheimer's disease

Emrin Horgusluoglu et al.

Summary: This study aims to identify AD-specific metabolomic changes and their potential upstream genetic and transcriptional regulators through an integrative systems biology framework. The findings suggest that short-chain acylcarnitines/amino acids and medium/long-chain acylcarnitines are most associated with AD clinical outcomes. Integration of gene expression data reveals ABCA1 and CPT1A are involved in regulating acylcarnitines and amino acids in AD.

Plasma metabolites were associated with spatial working memory in major depressive disorder

Yue Du et al.

Summary: The study identified abnormal metabolites in MDD patients using metabolomics, confirming a correlation between differential metabolites and neurocognitive dysfunction. MDD patients showed poorer performance in spatial working memory compared to healthy controls, with valeric acid being associated with working memory dysfunction in MDD individuals.

Solving Misclassification of the Credit Card Imbalance Problem Using Near Miss

Nhlakanipho Michael Mqadi et al.

Summary: This paper investigates the use of a hybrid data-point approach in resolving class misclassification issues in imbalanced credit card datasets, aiming to improve the effectiveness of machine learning algorithms in detecting fraud. The proposed method, combining feature selection with Near Miss-based undersampling, was tested on two different imbalanced credit card datasets, with random forest producing the best results among logistic regression, support vector machine, decision tree, and random forest algorithms.

Mechanistic basis for receptor-mediated pathological α-synuclein fibril cell-to-cell transmission in Parkinson's disease

Shengnan Zhang et al.

Summary: The study reveals that receptors LAG3 and APLP1 on cell surface can preferentially bind pathological alpha-synuclein in the amyloid state to facilitate cell-to-cell transmission in Parkinson's disease progression. Through biophysical experiments, it is found that LAG3 D1 and APLP1 E1 domains commonly use an alkaline surface to bind the acidic C terminus of alpha-synuclein, leading to increased binding affinity. Additionally, phosphorylation at serine 129 enhances the interaction between alpha-synuclein fibrils and the receptors, potentially contributing to the development of PD-like pathology.

Higher cerebrospinal fluid tau is associated with history of traumatic brain injury and reduced processing speed in Vietnam-era veterans: A Department of Defense Alzheimer's Disease Neuroimaging Initiative (DOD-ADNI) study

Alexandra L. Clark et al.

Summary: This study found that veterans with a history of traumatic brain injury had significantly higher levels of phosphorylated tau and total tau in CSF compared to those without a history of TBI. Additionally, TBI history moderated the association between CSF tau markers and cognitive functioning.
