Comparison of three synthetic transferrin mimetic small peptides to promote the blood-brain barrier penetration of vincristine liposomes for improved glioma targeted therapy

Solmaz Mojarad-Jabali et al.

Summary: The development of a modified liposomal formulation has shown promising results in facilitating brain targeted delivery and selective accumulation within glioma cells. Among the studied targeting ligands, T7-modified liposomes demonstrated superior BBB penetration, brain distribution, and increased accumulation in glioma cells, leading to improved anti-glioma outcomes through targeted cytotoxicity and apoptosis. These results suggest that T7-modified liposomes could serve as a potential platform for effective brain targeted delivery and glioma therapy in clinical settings.

Molecular architecture determines brain delivery of a transferrin receptor-targeted lysosomal enzyme

Annie Arguello et al.

Summary: The delivery of a lysosomal enzyme fused to a moderate-affinity transferrin receptor-directed blood-brain barrier enzyme transport vehicle results in superior brain delivery, biodistribution, and pharmacodynamics compared to a traditional high-affinity anti-TfR monoclonal antibody fusion. The differences in brain exposure, internalization, and substrate reduction of these two platforms highlight important features for the clinical development of TfR-targeting platforms in CNS diseases.

Shedding Light on the Blood-Brain Barrier Transport with Two-Photon Microscopy In Vivo

Krzysztof Kucharz et al.

Summary: Treatment of brain disorders depends on overcoming the blood-brain barrier, and Prof. Margareta Hammarlund-Udenaes's research has been instrumental in understanding the principles of drug delivery to the brain and developing related tools. This article introduces the use of in vivo 2-photon microscopy (2PM) to study the blood-brain barrier, focusing on characterizing the paracellular diffusion, adsorptive-mediated transcytosis, and receptor-mediated transcytosis of drug nanocarriers at the microscale.

IgG Fusion Proteins for Brain Delivery of Biologics via Blood-Brain Barrier Receptor-Mediated Transport

Ruben J. Boado

Summary: The treatment of neurological disorders requires the transport of therapeutic drugs across the blood-brain barrier (BBB). This can be achieved through the use of brain-penetrating bifunctional IgG fusion proteins, which consist of a transport domain and a therapeutic domain. These proteins target specific receptors on the BBB and have been validated in animal models and human clinical trials.

FOCUSED ULTRASOUND AND MICROBUBBLE TREATMENT INCREASES DELIVERY OF TRANSFERRIN RECEPTOR-TARGETING LIPOSOMES TO THE BRAIN

Marieke Olsman et al.

Summary: Research shows that combining FUS and microbubbles with BBB-targeting liposomes can enhance drug delivery to the brain. Transferrin receptor-targeting liposomes exhibited increased accumulation in the FUS-treated hemisphere, while isotype immunoglobulin G liposomes did not show significant increase.

Brain capillary pericytes and neurovascular coupling

Soren Grubb et al.

Summary: Neurovascular coupling is essential for matching cerebral activity with relevant blood flow, controlled by capillaries and precapillary arterioles. Mural cells, such as pericytes and smooth muscle cells, play a crucial role in determining blood vessel diameter and flow through their tone. The structure of these blood vessels and excitation-contraction coupling of mural cells are important factors to consider.

Post-capillary venules are the key locus for transcytosis-mediated brain delivery of therapeutic nanoparticles

Summary: Limited understanding of interactions between nanoparticle drug carriers and the blood-brain barrier underlies many translational failures in treatments of brain disorders. Here the authors use two-photon microscopy in mice to characterize the receptor-mediated transcytosis of nanoparticles at all steps of delivery from the blood to the brain in vivo.
