The underestimated contribution of the solvent to the phase behavior of highly drug loaded amorphous solid dispersions

Sien Dedroog et al.

Summary: The influence of organic solvents on the kinetic stabilization of drug components in a polymer matrix was investigated, indicating that the solvent determined the maximum amount of drug that could be kinetically trapped in the polymer matrix. There was no direct relationship found between the thermodynamic solubility of a drug and its highest drug loading attained using the same solvent, highlighting the important contribution of the solvent to the generation of a supersaturated state.

Infliximab formulation strategy for a stable ileo-colonic targeted oral dosage form intended for the topical treatment of inflammatory bowel disease

Bahez Gareb et al.

Summary: Research suggests that topically active infliximab therapy may be effective in treating IBD with fewer side effects. Oral administration of infliximab with site-specific delivery could be enabled through a sugar glass matrix. The stability and performance of IFX-I and ColoPulse-IFX were maintained during long-term storage, making them promising options for targeted treatment of IBD in the ileocolonic region.

Evaluation of lactose based 3D powder bed printed pharmaceutical drug product tablets

Korinde A. van den Heuvel et al.

Summary: Understanding the relationship between powder blend characteristics and tablet characteristics is crucial for obtaining 3D powder bed printed tablets that meet pharmaceutical specifications. This study focused on the impact of different particle size distributions on key material attributes such as wettability, consolidation, and flowability. Optimizing the base formulation and print settings concurrently is essential for successfully formulating hydrophilic/hydrophobic APIs into 3D printed tablets.

Ileo-Colon Targeting of the Poorly Water-Soluble Drug Celecoxib Using a pH-Dependent Coating in Combination with Self-Emulsifying Drug Delivery or Solid Dispersion Systems

Annemarie Broesder et al.

Summary: Targeting celecoxib to the ileo-colonic region using pH-dependent coating systems like ColoPulse may improve treatment and prevention of colon cancer. However, the dissolution behavior of celecoxib can be compromised due to its poor aqueous solubility, which can be addressed by combining pH-dependent coating with self-emulsifying drug delivery systems or solid dispersion systems.
