Enhanced Ocular Surface and Intraoral Nociception via a Transient Receptor Potential Vanilloid 1 Mechanism in a Rat Model of Obstructive Sleep Apnea

Summary: This study investigates the role of transient receptor potential vanilloid 1 (TRPV1) in mediating enhanced orofacial pain behavior and trigeminal spinal subnucleus caudalis (Vc) neuronal responses in a rat model of obstructive sleep apnea (OSA). The results show that chronic intermittent hypoxia (CIH) during the sleep phase enhances pain sensitivity in the ocular surface and intraoral mucosa through TRPV1-dependent mechanisms.

Evidence for a phenotypic switch in corneal afferents after lacrimal gland excision

Summary: This study investigated corneal innervation, ongoing pain, and alterations in corneal afferent phenotypes in a mouse model of severe aqueous tear deficiency. The results showed that dry eye-induced damage to corneal afferents led to corneal epithelial damage and ongoing ocular irritation. The corneal neurons underwent a regenerative process after injury, which was more significant in male animals. Additionally, dry eye also caused alterations in IB4-positive neurons, which may enhance neuroprotective mechanisms.

L-carnitine suppresses transient receptor potential vanilloid type 1 activity and myofibroblast transdifferentiation in human corneal keratocytes

Summary: Human corneal keratocytes (HCK) are essential for maintaining corneal structure and transparency. This study shows that the natural antioxidant L-carnitine inhibits stromal scarring by suppressing injury-induced intrinsic transient receptor potential vanilloid type 1 (TRPV1) activity that is linked to induction of myofibroblast transdifferentiation in HCK cells. Blocking TRPV1 activation on keratocytes may therefore be a viable approach to suppress corneal opacification in a clinical setting.

Summary: The study evaluated the effectiveness of TRPV1 blockade in alleviating ocular pain, neuroinflammation, and anxiety-like behaviors associated with severe DED. Results showed that TRPV1 antagonist reduced corneal responsiveness to various stimuli and inhibited the upregulation of genes involved in neuropathic and inflammatory pain in the TG, leading to reduced ocular pain sensation and anxiety-like behaviors in severe DED.
