Response characteristics of the membrane integrity and physiological activities of the mutant strain Y217 under exogenous butanol stress

Summary: The study shows that butanol inhibits bacterial activity by destroying the cell membrane and altering functionality of Clostridium acetobutylicum strains. Exogenous butanol led to changes in the carbon ion beam irradiation-mutant Y217 morphology. The mutant displayed lower microbial adhesion and intrinsically low membrane permeability. Additionally, DNA-level analysis revealed gene mutant sites related to membrane functions in the mutant strain Y217.

Butanol Tolerance of Lactiplantibacillus plantarum: A Transcriptome Study

Summary: The study identified genetic factors involved in the butanol stress response of Lactiplantibacillus plantarum, revealing common species-specific effects of butanol stress such as upregulation of multidrug efflux transporters, toxin-antitoxin system, and transcriptional regulators. The strains differed greatly in DEGs responsible for membrane transport, metabolism, and important transcriptional regulators, with uniquely upregulated genes in the butanol-resistant strain contributing to tolerance. The phosphoenolpyruvate flux and the phosphotransferase system were also found to be major factors in butanol tolerance.

Genetic modification of Clostridium species by introducing stress tolerance-associated genes may be a promising strategy for improving butanol tolerance and production.

Summary: Engineering synthetic scaffoldins derived from C. cellulovorans cellulosome components successfully introduced them into L. lactis, with a focus on their potential cell-surface anchoring capabilities. Among the engineered scaffoldins, only those carrying three EngE SLH modules were able to bind to the surface of L. lactis, despite the absence of the conserved TRAE motif typically involved in binding with secondary cell wall polysaccharides. These synthetic scaffoldins could potentially be used for assembling secreted or surface-displayed designer cellulosomes in L. lactis.
