Tebentafusp in advanced uveal melanoma: proof of principle for the efficacy of T-cell receptor therapeutics and bispecifics in solid tumors

Alex W. Liu et al.

Summary: Tebentafusp is a novel bispecific immune mobilizing TCR-based agent developed for metastatic uveal melanoma, showing potential to become the standard of care. The review discusses preclinical studies, clinical trials, and phase III trial results demonstrating improved overall survival compared to standard of care. Future studies should focus on identifying biomarkers for treatment response or toxicity prediction, as well as exploring combination therapy and adjuvant use of tebentafusp to reduce recurrence risk.

New targeted and epigenetic therapeutic strategies for the treatment of uveal melanoma

Alexander Z. Wei et al.

Summary: Uveal melanoma (UM) is a rare ocular malignancy with limited treatment options for metastatic disease. Efforts are being made to understand the disease-specific mechanisms and identify potential targets for therapy. Preclinical studies and clinical trials are being conducted to develop targeted therapies for UM.

Incidence and characteristics of metastatic intracranial lesions in stage III and IV melanoma: a single institute retrospective analysis

Mani Ratnesh S. Sandhu et al.

Summary: The study found that 22.9% of patients with stage III melanoma and 37.7% of patients with stage IV melanoma developed BM. The incidence of BM development significantly decreased from 2012 to 2017. Patients with BM had a significantly higher frequency of mutational BRAF in their primary tumor.

Overall Survival Benefit with Tebentafusp in Metastatic Uveal Melanoma

Paul Nathan et al.

Summary: The randomized trial of tebentafusp for metastatic uveal melanoma showed a higher 1-year overall survival rate among patients who received tebentafusp compared to those who received the investigator's choice of therapy, indicating superior efficacy.

Metastatic Risk Factors Associated with Class 1A Uveal Melanoma Patients

Alexej Ballhausen et al.

Summary: The study found that in uveal melanoma Class 1A patients, those with stage III disease and/or large COMS size are at elevated risk for metastasis. Combined clinical decision-making using AJCC stage and COMS size can have a significant impact on improving risk stratification and adapting follow-up intervals for Class 1A UM patients.
