CC-01 (chidamide plus celecoxib) modifies the tumor immune microenvironment and reduces tumor progression combined with immune checkpoint inhibitor

Summary: This study evaluated the combination of chidamide and celecoxib with PD-1 inhibitors in regulating the tumor microenvironment. The combination therapy significantly reduced tumor size, increased treatment efficiency and survival rate. Immune analysis revealed a decrease in immunosuppressive cells and activation of immune cells in the tumor microenvironment. Furthermore, gene analysis showed upregulation of genes associated with interferon-gamma response. Therefore, this combination therapy is important for enhancing the anti-tumor effect and immune response rate.

This study found that TREM-1 and TREM-2 are co-expressed on tumor-infiltrating myeloid cells, and elevated sTREM-1 is associated with disease outcomes. The results suggest that modulating the balance of TREM-1/TREM-2 may be a novel means to modulate the phenotype and function of tumor-infiltrating myeloid cells.

Summary: This study reveals that C5a can enhance the capacity of PMN-MDSCs to promote tumor growth and metastasis by inducing the formation of NETs. The formation of NETs is dependent on the production of HMGB1 by cancer cells. Inhibiting C5a, C5aR1, or NETosis can reduce the number of circulating tumor cells and the metastatic burden in a mouse lung metastasis model. The translational relevance of these findings is supported by the stimulation of migration and NETosis in PMN-MDSCs obtained from lung cancer patients.
