High Fat Diet Multigenerationally Affects Hippocampal Neural Stem Cell Proliferation via Epigenetic Mechanisms

Neural Stem Cell-Derived Extracellular Vesicles Counteract Insulin Resistance-Induced Senescence of Neurogenic Niche

Summary: The depletion of neural stem and progenitor cells (NSPCs) may contribute to cognitive impairment in age-related diseases, and insulin resistance has been linked to brain dysfunction. In an experimental model of insulin-resistant NSPCs, researchers identified a molecular circuit involving insulin receptor substrate-1 (IRS-1)/Forkhead box O (FoxO) signaling cascade that inhibits the activation of transcription factors FoxO1 and FoxO3a, which regulate cell proliferation and self-renewal. Insulin resistance also increased the expression of cyclin-dependent kinase inhibitor 1 (p21) and accelerated NSPC senescence. However, stimulation of NSPCs with NSPC-derived exosomes (exo-NSPC) rescued IRS-1/FoxO activation and countered the reduced proliferation and senescence of stem cells. Administration of exo-NSPC via the nasal route also restored the balance between proliferating and senescent NSPCs in the hippocampus of mice on a high-fat diet, improving adult hippocampal neurogenesis. These findings provide insights into the metabolic control of NSPC fate and the role of extracellular vesicles in regulating cell fate in the adult neurogenic niche.
