Human alveolar type 2 epithelium transdifferentiates into metaplastic KRT5+ basal cells

Jaymin J. Kathiriya et al.

Summary: The study reveals that human alveolar type 2 cells can transdifferentiate into basal cells in response to fibrotic signaling, driving the pathology associated with severe lung injuries. Aberrant mesenchyme plays a crucial role in the lineage trajectory leading to the expansion of alveolar metaplastic basal cells.

Pseudohypoxic HIF pathway activation dysregulates collagen structure-function in human lung fibrosis

Christopher J. Brereton et al.

Summary: This study provides evidence that the activation of the hypoxia-inducible factor (HIF) pathway is a critical regulator of pathogenetic collagen structure-function in fibrosis. Oxidative stress can drive the activation of pseudohypoxic HIF pathway, which alters collagen nanostructure and increases tissue stiffness.

Intermittent hypoxia increases ROS/HIF-1α 'related oxidative stress and inflammation and worsens bleomycin-induced pulmonary fibrosis in adult male C57BL/6J mice

Mengqing Xiong et al.

Summary: The study revealed that intermittent hypoxia exacerbated BLM-induced pulmonary fibrosis, with higher expression of ROS and HIF-1 alpha related to oxidative stress and inflammation. There was a positive linear correlation between HIF-1 alpha expression and hydroxyproline content, as well as a positive correlation between inflammation and ROS expression.

Hypoxia in Cancer and Fibrosis: Part of the Problem and Part of the Solution

Yair Romero et al.

Summary: Adaptive responses to hypoxia play crucial roles in the progression of lung cancer and pulmonary fibrosis, with hypoxia potentially serving as the link between these diseases. The key role of hypoxia-inducible factors (HIFs) and interconnected feedback loops contribute to HIF overactivation, making it an irreversible phenomenon. Further research may identify new opportunities in therapy and regeneration by focusing on HIF signaling and its potential overlap in these diseases.

Mild chronic hypoxia-induced HIF-2α interacts with c-MYC through competition with HIF-1α to induce hepatocellular carcinoma cell proliferation

Han Mu et al.

Summary: This study aims to investigate the balance between HIF-1 alpha and HIF-2 alpha in hypoxic tumor growth, particularly in HCC. The results revealed that HIF-2 alpha expression positively correlated with c-MYC in tumor tissues, and under mild chronic hypoxia, HIF-2 alpha competed with HIF-1 alpha to bind to c-MYC. Targeting the PI3K/mTORC2/HIF-2 alpha/c-MYC axis may provide a potential treatment option for HCC patients, especially those with rapidly growing tumors under mild chronic hypoxic conditions.
