Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Hypoxia-induced long noncoding RNA NR2F1-AS1 maintains pancreatic cancer proliferation, migration, and invasion by activating the NR2F1/AKT/mTOR axis

Yanqing Liu et al.

Summary: This study investigated the biological effects and regulatory mechanisms of NR2F1-AS1 in pancreatic cancer. The overexpression of NR2F1-AS1 was found to be correlated with poor prognosis and aggressive behavior in pancreatic cancer. Depletion of NR2F1-AS1 inhibited cell proliferation, migration, invasion, and tumor growth and metastasis in vitro and in vivo. Mechanistically, NR2F1-AS1 positively regulated the neighboring NR2F1 gene, leading to the activation of AKT/mTOR signaling and upregulation of HIF-1 alpha. Further investigations also revealed that NR2F1-AS1 expression was transcriptionally regulated by HIF-1 alpha under hypoxic conditions.

The Role of Cancer-Associated Fibroblasts in Ovarian Cancer

Mo Zhang et al.

Summary: This review focuses on the important role of cancer-associated fibroblasts (CAFs) in the ovarian cancer tumor microenvironment. Different subtypes of CAFs have specific functions in tumor pathogenesis and can be potential treatment targets. Several clinical or preclinical trials have targeted CAFs to enhance ovarian cancer treatment outcomes.

NR2F1, a Tumor Dormancy Marker, Is Expressed Predominantly in Cancer-Associated Fibroblasts and Is Associated with Suppressed Breast Cancer Cell Proliferation

Rongrong Wu et al.

Summary: Tumor dormancy is an important mechanism for late breast cancer recurrence. This study investigates the clinical relevance of NR2F1 expression, a known biomarker for dormancy. The results show that high NR2F1 expression in breast cancer is associated with enriched metastasis and stem cell-related pathways, as well as suppressed cell proliferation. Furthermore, NR2F1 is predominantly expressed in cancer-associated fibroblasts, rather than in cancer cells.

Targeting the tumour microenvironment in platinum-resistant ovarian cancer

M. Cummings et al.

Summary: Ovarian cancer often presents at an advanced stage and initial response to platinum-based therapies is positive, but chemoresistance develops leading to poor prognosis. The tumor microenvironment has been found to play a central role in therapy resistance and disease progression, making it an attractive therapeutic target. Focus has been on targeting cancer associated fibroblasts, tumor-associated macrophages, cancer stem cells, and cancer cell metabolic vulnerabilities for new therapies.

Homologous Recombination Deficiency Testing for BRCA-Like Tumors: The Road to Clinical Validation

Marjolijn M. Ladan et al.

Summary: Effective cancer treatment requires selecting patients based on tumor characteristics, including those with BRCA1/2 mutations or defects in the homologous recombination repair pathway. The challenge lies in identifying patients with BRCA-like tumors and selecting appropriate treatment strategies. Germline BRCA mutations result in HRD phenotype, making tumors sensitive to DNA double strand break-inducing agents.

Platinum Complexes in Colorectal Cancer and Other Solid Tumors

Beate Koeberle et al.

Summary: Cisplatin is effective in treating various solid tumors, but not in colorectal cancer due to alterations in DNA damage signaling and repair mechanisms. Improvement in cisplatin-based chemotherapy for colorectal cancer may be achieved through newly designed analogues and biochemical modulators. Overcoming resistance in cancers can involve targeting DNA damage signaling pathways and utilizing improved platinum compounds.

Summary: This study reveals the regulatory role of lncRNA NR2F1-AS1 in metastatic dormancy, promoting tumor dissemination but inhibiting proliferation in mesenchymal-like breast cancer stem-like cells. This connection between BCSC plasticity and metastatic dormancy highlights the importance of lncRNA in these processes.

Mutated p53 in HGSC-From a Common Mutation to a Target for Therapy

Aya Saleh et al.

Summary: Mutations in the tumor suppressor gene TP53, encoding for the p53 protein, are common in ovarian high-grade serous cancer (HGSC), leading to loss of tumor suppressive function and gain of new oncogenic functions. This review highlights the clinical relevance of TP53 mutations in HGSC, including upstream regulators of mutant p53, downstream target genes, and p53's role in HGSC pathogenesis. It also discusses the potential of TP53 mutations as diagnostic biomarkers and therapeutic targets in HGSC.

Deciphering the Collagen code in tumor progression

Archana Anshuman Gupta et al.

Summary: Cancer is considered a disease of imbalance, influenced by both genetic and epigenetic mechanisms. The tumor microenvironment, particularly the extracellular matrix (ECM), plays a crucial role in tumor progression through its interaction with tumor cells. Recent studies have highlighted the biochemical and biophysical signaling properties of ECM, especially collagen, implicating it in the promotion of tumor progression and metastasis.

Targeting TGFβ signal transduction for cancer therapy

Sijia Liu et al.

Summary: TGF beta family members are cytokines with diverse effects on cell fate regulation during development and tissue homeostasis. Dysregulation of TGF beta signaling can lead to developmental disorders and diseases, including cancer. TGF beta plays a dichotomous role in cancer progression, acting as a tumor suppressor in early stages and a tumor promoter in late stages, with effects on proliferation, differentiation, apoptosis, metastasis, and the tumor microenvironment.
