Proliferative Potential, and Inflammatory Tumor Microenvironment in Meningioma Correlate with Neurological Function at Presentation and Anatomical Location-From Convexity to Skull Base and Spine

Johannes Wach et al.

Summary: The anatomical location of meningiomas may be associated with the prognosis, and there are differences in survival, proliferative potential, and density of tumor-associated macrophages between spinal and skull base meningiomas.

Inflammatory Tumor Microenvironment in Cranial Meningiomas: Clinical Implications and Intraindividual Reproducibility

Johannes Wach et al.

Summary: This study compares the MIB-1 index of meningiomas, which shows strong intraindividual reproducibility and a direct correlation with time to recurrence. Individual MIB-1 indices can be used for tailored follow-up imaging intervals. Inflammatory factors and macrophages may play an important role in the regrowth potential of meningiomas.

Predictors of postoperative seizure outcome in supratentorial meningioma

Ron Gadot et al.

Summary: This study retrospectively analyzed the medical records of 384 patients who underwent meningioma resection from 2008 to 2020, finding that preoperative seizures have an impact on postoperative seizure outcomes, with distinct margins on preoperative imaging, Simpson grade I resection, postresection ischemia, WHO grade, and recurrent disease being associated.

High Mib-1-score correlates with new cranial nerve deficits after surgery for frontal skull base meningioma

Matthias Schneider et al.

Summary: This study aimed to analyze various factors influencing cranial nerve function in frontobasal meningioma surgery, finding that elevated Mib-1 labeling indices were associated with immediate postoperative new cranial nerve dysfunction. The study suggests that macrophage-based inflammatory immune responses may play a role in the occurrence of postoperative cranial nerve deficits.

Sex-Related Differences in Drugs with Anti-Inflammatory Properties

Andre Farkouh et al.

Summary: There is increasing evidence of sex differences in the action of anti-inflammatory drugs, with women being at significantly higher risk of adverse effects. Men tend to have stronger anti-thrombotic activity with COX-inhibitors, while women are more sensitive to corticosteroids when their estradiol levels are high. Research on sex differences in drug effectiveness and safety is growing, but findings are often anecdotal and controversial, with a lack of systematic sex-differentiated reporting in clinical trials.

Evaluating the impact of age on immune checkpoint therapy biomarkers

Rossin Erbe et al.

Summary: Both tumors and aging can alter the immune landscape of tissues, affecting the efficacy of immune checkpoint blockade (ICB) therapy in elderly patients. While age-related changes may enhance ICB efficacy in older patients, such as increased tumor mutational burden and interferon gamma signaling, there are also alterations that could reduce ICB efficacy, like decreases in T cell receptor diversity. Further large-scale prospective studies on ICB therapies among elderly patients are warranted to fully understand these effects.

FORGE: A Novel Scoring System to Predict the MIB-1 Labeling Index in Intracranial Meningiomas

Johannes Wach et al.

Summary: This study identified baseline variables associated with the MIB-1 labeling index in meningiomas, developed a scoring system to estimate the risk of an elevated MIB-1 index before surgical resection, and found that this score may predict progression-free survival. The study suggests that the novel score may help guide preoperative surgical planning, postoperative follow-up scheduling, and future trials investigating inflammatory burden and proliferative activity in meningioma patients.
